線粒體OCR探針
線粒體OCR試劑是一種基于最新納米技術(shù)研制的可主動靶向活細(xì)胞線粒體的成像試劑。該試劑生物相容性好,細(xì)胞毒性很低,可廣泛應(yīng)用于單細(xì)胞、細(xì)胞球Spheroids或者組織類器官等應(yīng)用場景,實現(xiàn)細(xì)胞中氧氣含量的測量。本公司所研制的線粒體OCR試劑具有優(yōu)異的光穩(wěn)定性,可通過熒光壽命測量技術(shù)實現(xiàn)細(xì)胞中氧濃度變化的實時跟蹤,同時也能通過測量密封環(huán)境中細(xì)胞的氧氣消耗情況,計算細(xì)胞的耗氧速率OCR,從而實現(xiàn)對細(xì)胞線粒體功能的實時評估。如中圖所示,線粒體OCR試劑的Zeta電位為正,保證了試劑可以有效地逃離溶酶體,準(zhǔn)確靶向到細(xì)胞線粒體上。同時試劑具有優(yōu)異的光穩(wěn)定性,在使用門控技術(shù)進行熒光壽命測量時,其可以穩(wěn)定、可靠并且準(zhǔn)確地測量細(xì)胞內(nèi)氧濃度的動態(tài)變化,連續(xù)測量時間可長達數(shù)小時,如右圖所示。
The mitochondrial oxygen consumption rate (OCR) reagent is an imaging reagent based on the latest nanotechnology that can actively target the mitochondria of living cells. The reagent has good biocompatibility and low cytotoxicity, and can be widely used in applications such as studying energy metabolism of single cells, cell spheroids or tissue organoids. The mitochondrial OCR reagent developed by our company has excellent photostability, and can achieve real-time tracking of oxygen concentration changes in living cells through fluorescence lifetime imaging technology. It can also calculate the oxygen consumption rate of living cells by measuring their oxygen consumption in a sealed environment, thus achieving real-time evaluation of the function of cellular mitochondria. As shown in the middle figure, the Zeta potential of the mitochondrial OCR reagent is positive, ensuring that the reagent can effectively escape from the lysosome and accurately target the mitochondria. At the same time, the reagent has excellent photostability, and when using gating technology for fluorescence lifetime measurement, it can stably, reliably, and accurately measure the dynamic changes in intracellular oxygen concentration, with continuous measurement time up to several hours, as shown in the right figure.
線粒體OCR試劑具有完全可逆的氧氣響應(yīng)行為,不僅可以通過熒光強度測量,而且可以通過測量熒光壽命準(zhǔn)確定量線粒體中的氧濃度,該試劑完全兼容現(xiàn)有的時間分辨酶標(biāo)儀設(shè)備,不需要額外的測量系統(tǒng)即可實現(xiàn)測量。試劑具有優(yōu)異的響應(yīng)靈敏度和響應(yīng)速度,可在不到1s的時間內(nèi)實現(xiàn)氧濃度的精準(zhǔn)定量測量。
The mitochondrial OCR reagent has a completely reversible oxygen response behavior, which can be accurately quantified not only by measuring fluorescence intensity, but also by measuring fluorescence lifetime. This reagent is fully compatible with existing time-resolved microplate reader equipment for high throughput measurement. The reagent has excellent response sensitivity and response speed, and can achieve accurate quantitative measurement of oxygen concentration in less than a second.
線粒體OCR試劑不僅適用于高通量的96孔板進行細(xì)胞、藥物和培養(yǎng)條件的篩選,也能進行顯微成像,實現(xiàn)單/活細(xì)胞中氧濃度的測量,對類器官、組織切片也有很好的適用性。
The mitochondrial OCR reagent is not only suitable for high-throughput 96 well plate screening of cells, drugs, and culture conditions, but also for microscopic imaging, achieving measurement of oxygen concentration in single/live cells, and has good applicability to organoids and tissue sections.
